Enzymatic activity among other things...

This (blogging) is my short escape from writing my lab report that I have been sitting here trying to make sense of for about 5 straight hours. That and the online bingo game that I play once an hour just so that my brain won’t frizzle and explode into small pieces resembling the ferrochelatase protein that I am writing about. Aaaahhhh!


Here is a sample of the lab report I am writing, just so you know what I am going through…

Elueringsbuffert: Totalt behövdes 30 ml
Vildtyp: 15 ml
Mutant: 15 ml
1) För att få 4x spädning:
1x + 4x = 30 ml
x = 6 ml
Så 6 ml elueringsbuffert + 24 ml vatten = 4x elueringsbuffert
2) Spädes med avjonat vatten till 1x:
C1V1 = C2V2
(4x)(V1) = (1x)(30 ml)
V1 = 7.5 ml bindningsbuffert, spädes upp till 30 ml med vatten.
Diskussion av SDS-PAGE gel
Molekylarviktsstandarden hjälper till med att kunna identifiera närvaron av ferrokelatas i proverna. Sträckan vid 45000 Da i standarden ska kunna jämföras med sträckan i proverna där ferrokelatas har en vandringssträcka som motsvarar en molekylvikt på 40,5 kDa.
Det syns tydligt i både vildtyp och mutant supernatanterna att det finns många sträckor som tyder på ett orent prov. Ett mörkt band syns dock tydligt vid 45000 Da vilket indikerar närvaron av ferrokelataset.
En sträcka där ferrokelatas ska finnas kan också ses i både vildtyp avsaltat och mutant avsaltad. Här är det nästan bara det bandet som syns, precis så som det ska vara eftersom de avsaltade proverna ska innehålla endast ferrokelatas.
Det syns ingenting längs tvättfraktionerna för både vildtyp och mutant, vilket var förväntat.
Flera sträckor syns längs bindningsfraktionerna men även här är ferrokelatas sträckan tydligast.
Okay, okay enough of that. But, I should be done in a few hours. Then all I have to do for this class is study for the big mega exam that is coming up on June second. And if I pass that I get two weeks off to do cool hobby stuff (like making videos and rollerblading) until we leave for the USofA!
Yesterday we had a lovely sunset. It would have been perfect to drive out to the west coast (about 15 minutes' drive) to watch the zillions of ever-changing hues as the sun slowly made its way down into the horizon. But, I watched it from the computer desk instead since I was writing my lab report. But at least I could see it from the window!
All this weekend Niklas' family was here for a visit. On Saturday we took the car across the new bridge to Copenhagen and spent a few hours there. Copenhagen is about as multi-cultural as you can get. It was like Disney World in that you could hear about 17 different languages being spoken all at the same time. It was a lovely late spring evening and since the sun doesn't set until like 9:45 here it was very pleasant.

Latest webpage visited: Kunle and Sarah's Blog...there was a new entry about a grinding mill.
Weather right now: CHILLY and not at all like the second half of May should be.
Reading: an interesting novel by a Christian author called "Merlin" and 2 Corinthians.
Last movie I saw that was any good: a movie called "I am David" that I can recommend because it is really clean and has a good message.
Highlight of the past weekend: Rollerblading with Niklas. I am getting better and better and I am thankful that he is so patient with me! (he is really good)
Something that has made me thankful the past few days: thinking about the first phase of my relationship with Niklas (how we fell in love and became a couple). I was thinking about it recently because I read my journal from that time last night before going to bed.
Worship CDs that I highly recommend but don't own yet: the new stuff by Terry McAlmon.
One of the things I look forward to about the 5 weeks in the U.S. : warm summer evenings and lightning bugs and the smell of blooming flowers and trees at night, as well as driving with the windows down.
Ok, back to enzymatic activity and purification factors.

  1. You know, Katie, it is amaziing. I can say with confidence that I know you pretty well. We have spent some quaility time together and we are honest and forthright with each other. But after reading the equations you are working on you must be a complete stranger. That just boggled my mind!
